Studies on the Homogenate ATPase Activity of the Rat Brain Cortex

Abstract
The activity of Mg activated and Na-K activated ATPase of homogenate from rat brain cortex was measured in vitro under the variety of conditions.
The effect of general depressants such as acetone, chloroform, ethanol, pentanol, octanol, ethyl ether and penthrane on the activities of both ATPase was investigated and the results were summarized as follows.
1. The homogenate contained Na+ plus K+ activated ATPase in the presence of Mg++. The Na-KATPase activity increased as the concentration of protein increases.
2. The Na-K ATPase activated by monovalent cations such as NH,+ and Li+ other than K+.
3. In the presence of lower level of K+ or NH,+, the inhibition produced by ethanol on the Na-KATPase increased as the concentration of Na+ of the incubation medium was raised.
4. Na-K ATPase was more markedly inhibited than the Mg ATPase by acetone, chloroform, ethanol and pentanol.
5. Both Mg and Na-K ATPase activities were markedly inhibited by octanol, ethyl ether and penthrane.
6. The inhibition of Na -K ATPase produced by acetone, octanol and ethyl ether except penthrane was higher at 100 mM than at 25 mM Na+.
These finding indicate that the nature of ATPase isolated from the homogenate of rat brain cortex was quite similar to that from other organs, and then remarkable correlation between anesthetic potency and ATPase inhibition could thus be observed.
